ARBOVIRUS ISOLATIONS

Methods. Mosquitoes were sorted into pools of £25 mosquitoes, according to locality, site, date of capture, species, sex and (for females) whether blood-engorged or not. Each mosquito pool was then ground (Doggett et al., 1997), centrifuged and the supernatant inoculated onto various cell lines (C6/36, BHK and PSEK cells). The C6/36 plates from the first inoculation were screened for the presence of flaviviruses via a 'fixed cell' enzyme linked immunosorbent assay (ELISA)(Broom et al., 1998). This assay is also used to identify any suspected viral isolate, and can identify the alphaviruses - Barmah Forest (BF), RR, SIN and Getah (GET), and the flaviviruses - MVE, KUN, Alfuy (ALF), EH, KOK and STR.  Any isolate that was not identified by this assay was labelled as 'unknown'.

All positive virus results were sent to Dr Jeremy McAnulty of the AIDS and Infectious Disease Branch, NSW Health, and to the relevant Public Health Unit. Virus results were also posted on the NSW Arbovirus Surveillance Web Site (under ‘Mosquito/Chicken Results’) and under each location's results.


Results. Of the 245,524 mosquitoes processed, there were 33 viral isolates. These are listed in the tables below.
